This set of three seed stocks and two DNA stocks can be used with the "Gibberellic Acid Physiology and Plant Transformation" module to introduce GA (gibberellic acid) hormone physiology and plant transformation in an undergraduate laboratory module. In this module, Arabidopsis ga5 semi-dwarf mutant plants with reduced activity of GA 20-oxidase are transformed with a construct containing the GA5 gene. The dwarf mutant plants transformed with a wild-type version of the gene show normal size, demonstrating successful complementation of the mutant phenotype. Besides visual observation of the phenotype, students learn additional techniques, including PCR and agarose gel electrophoresis. This experiment is cost effective and can be completed within a 4-month term.
